Monkey pox is a zoonotic virus, causing symptoms similar to small pox, though less severe clinically. It belongs to orthopoxvirus genus of Poxviridae family. Two clades of Monkeypox are known: the West African and the Congo Basin (Central African). It can be transmitted from one person to other through close contact with lesions, body fluids, droplets and contaminated material such as linen. The incubation period for infection is usually 6-13 days, however it can vary from 5 to 21 days. As per WHO, possible symptoms may include headache, fever, swollen lymph nodes, weakness, muscle and body ache. The Monkeypox virus can be detected by real time polymerase chain reaction, transmission electron microscopy and immunohistochemistry.
The Monkeypox Virus Real Time PCR kit is used for the detection of Monkeypox virus in serum or lesion exudate samples by using real time PCR systems
1. LOD: The limit of detection is 200 copies/ml.
2. Precision: Coefficient of variation (CV%) of precision Ct value within batch ≤3%.
3. Specificity: There is no cross reaction between the kit and positive samples, such as Smallpox Virus, Cowpox Virus,
Vaccinia Virus etc.
1. Reagent Preparation(performed at "reagent preparation region")
1.1 Take out each componentsfrom the diagnostic kitand placethemat room temperature. Allow thereagents to equilibrateatroom temperature, then vortex each of themrespectively forlater use.
1.2 According to thequantityof test specimens, MPV Positive Control, MPVNegative Control,and Internal control (IC), pipetteappropriate quantity of MPVPCR Mix andMPVEnzymeMix ( MPV PCR Mix 15 μl/test +MPV EnzymeMix 4 μl/test +Internal control (IC) 1 μl/test), mix them thoroughlytomake a PCR-Mastermix, centrifuge itinstantaneously forlater use.
1.3 Transfer the above-prepared reagents to the"specimen processing region" for later use.
2. Processing and loading ofspecimens (performed at "specimenprocessing region")
2.1 Thisdiagnostic kit does not include Viral RNA&DNA Extraction Kit. Itisrecommended to use Viral RNA&DNA ExtractionKit produced by Changsha Renji Medical Equipment Co., Ltd. to extract viral DNA. The specific operation is in accordancewith its instructions.
2.2 Add 20μl PCR-Mastermix into PCR reaction tube with 5μl above processed sample, MPV Positive Control and MPV NegativeControl, and cap the tube. Carry outfluorescence quantitativePCR detection on fluorescencePCR instrument.
3.PCRAmplification (performed at "nucleic acidamplificationarea")
3.1 Place PCRreaction tubes into thespecimenwells of the amplification equipment. SetupMPVPositive Control, MPV NegativeControl and specimens to be tested in the corresponding sequence and input specimen name.
3.2 Set cycle parameters according to the following table for PCR amplification.
